Stable Isotopic Labeled Mice and Mice Tissue

 

Silantes GmbH provides metabolically labeled mice and mice tissue using the labeled mice diets produced and developed in-house.

Labeled mice or mice tissue are useful as standards for quantification of proteomic data. Fig. 1 shows the work flow.

In order to quantify differences in protein patterns of mouse (or organ) A with respect to mouse (or organ B), the isotopically labeled mouse (organ) R is mixed with A and B. The proteomes of sample A mixed with R (in a 1:1 ratio)  and B mixed with R (in 1:1 ratio) are isolated, digested and subjected to LC-MS as shown in the work flow (Fig.1). Two ratios are determined: the ratio of peptide amounts stemming from A and R is determined yielding the (A/R)-ratio and the (B/R)-ratio for each peptide. Calculating the ratio (A/R): (B/R) yields the ratio of peptides (A/B).

It is worth mentioning that the strain for R must not necessarily be the same as that used for A and B. The stable isotopically labeled mouse or organ (R) is used as a standard, permitting normalising A with respect to B, accounting for differences in the isolation procedure in the culture mixtures (A/R) and (B/R) without affecting the peptide ratios (A/B).

In other words, the customer is free as regards the choice of strain used for the experiment, but can use the Silantes-labeled mouse (or organ) as a reference which is spiked into tissue A and B.

Mice and organs with the following labels are available:

-          13C, lysine specific labeled                  (Lys(6)-SILAC)

Mouse strain (R) is C57 BL 6.
